Antibody-antigen binding may be monitored on the SPRimager II either by (i) immobilizing antigen (or peptide eitopes) on gold chips, then exposing them to antibody solution. In this example, biotin-labelled antibodies are immobilized on streptavidin-coated SpotReady chips. Specific binding of IFN antigen to anti-IFN antibody is demostrated, confirming that immobilized antibody is active. A biotinylated oligonucleotide ligan binds only to control streptavidin spots with no antibody attached, confirming the functional activity of the streptaviin surface and the specifity of the antigen binding.
