资料下载心理应激动物模型的制作与行为学测试 一、心理应激动物模型的制作 人们在复制心理应激动物模型方面做了大量工作,但情绪毕竟是复杂的,躯体应激与心理应激因素的区别始终是应激研究中的难关,难于绝对地在实验中进行模拟或类比。下面是几种常见的动物模型制作方法,为进行心理应激研究提供参考。 1 . 社交失败应激动物模型: 雄性大鼠放入单独饲养大鼠笼中, 单独饲养大鼠会攻击实验组大鼠, 撕咬侵入者的头部、颈部和背部(无外伤发生)。社交失败的定义是:实验大鼠至少受到一次攻击, 并且表现出驯服的姿势, 如防御性直立、仰 卧、静止不动等 。由于最后得到的实验对象未受到明显的躯体刺激, 因而可以认为是较理想的心理应激模型。 2 .Communication Box 大鼠模型: 被电击的大鼠惊叫、惊跳; 实验组大鼠不受电击, 但旁观其他大鼠遭受电击的过程,通过视觉、听觉等而产生心理应激。每次持续30分钟,隔天1次, 连续两周 。 3 . 行为限制( 束缚制动) : 将造模大鼠置于自制束缚制动盒内, 限制大鼠的活动空间, 每日1 次, 每次2小时,造模大鼠在制动期间禁食、禁水。6天后进行检测。 4 . 多重复合慢性应激: 将隔离应激、新环境应激、拥挤、社交失败应激等4 种刺激随机安排到28 日内,每日1种,产生慢性轻度不可预见性的应激。由于慢性应激是模拟抑郁症的环境诱因,动物的行为特征改变、血浆皮质激素升高等均与内源性抑郁症状相似,作为抑郁模型具有较高的价值。
