Description
5-Methylcytosine is the methylated form of the DNA base cytosine. In mammalian cells, DNA methylation is an enzymatic modification at the 5-position of cytosine present abundantly within the CpG dinucleotide sequence context. This DNA modification is inheritable and reversible without primary DNA base sequence changes resulting in possible epigenetic modulation of phenotype and gene expression (1). CpG methylation may directly interfere with the binding of certain transcriptional regulators to their cognate DNA sequences or may enable recruitment of methyl-CpG binding proteins that create a repressed chromatin environment (2). The de novo formation and maintenance of 5-methylcytosine is catalyzed by DNA methyltransferase proteins (DNMTs) (1).

MeDIP: 4 ug of sonicated Jurkat genomic DNA or 0.5 ug of fully methylated Jurkat DNA (Active Motif) were immunoprecipitated with a rabbit IgG or a RabMAb against 5-methylcytosine (cat.# 3913-1). 2 ul of immunoprecipitated DNA elute or 50 ng of fully methylated Jurkat DNA together with BRCA1 primer mix were used for PCR.

2,3-Jurkat genomic DNA (4 ug) for IP
4-fully methylated Jurkat DNA (0.5 ug) for IP
5-fully methylated Jurkat DNA (50 ng) Competitive ELISA: 0.1 ug/ml of BSA-methylcytosine was coated into 96-wells. 5 mg/ml of competitor's methylcytosine, cytosine, or hydroxymethylcytosine and 0.1 ug/ml of anti-5-Methylcytosine antibody (cat.# 3913-1) were added. HRP conjugated goat anti-rabbit IgG antibody was used to develop the color.

Specificity
KLH-5-Methylcytosine was used as immunization.

Storage Condition and Buffer
Store at -20° C. Buffer: 50 mM Tris-Glycine (pH 7.4), 0.15 M NaCl, 40% Glycerol, 0.01% sodium azide and 0.05% BSA. Stable for 12 months from date of receipt.
