A DNA sequence encoding the SARS-CoV-2 Spike S1 (YP_009724390.1, with mutations (157-158) deletion, T19R, G142D, E156G, L452R, T478K, D614G, P681R) (Met1-Arg685) was expressed with a c-terminal polyhistidine tagged AVI tag at the C-terminus. The purified protein was biotinylated in vitro. The mutations were identified in the SARS-CoV-2 variant (known as variant B.1.617.2) which emerged in the India.
分子量
The recombinant SARS-CoV-2 Spike S1 consists of 694 amino acids and predicts a molecular mass of 78.09 kDa. As a result of glycosylation, it migrates as an approximately 85.61 kDa band in SDS-PAGE under reducing conditions.
