新型数据非依赖采集质谱技术及其生物学应用 Advanced Data-independent Acquisition Mass Spectrometry for Biological Research

Biological samples frequently generate a complex analytical matrix. Recent innovative DIA-MS approaches in both fast-scanning mass spectrometer (MS) such as Q-TOF and high-resolution Orbitrap MS provide advanced analytical capacities. The current DIA methods mostly have to use rather large isolation windows, resulting in multiplexed MS2 spectra that cannot be directly analyzed by shotgun database searching engines. On the other hand, the bottom-up proteomics has common experimental features and principles, such as the similar peptide length, similar peptide charge distributions, the same sequential elution of the peptides along LC separation, and the predictable mass-to-charge (m/z) difference between peptides and their post-translational modification (PTM) forms. Based on the above common features of the bottom-up proteomic workflows, we will discuss how we devised novel and intelligent DIA strategies and bioinformatic solutions facilitating systems biological research.
