相干拉曼应用于代谢产物和特定蛋白的化学成像

Super-resolution (SR) optical microscopy is a dramatic advance that breaks diffraction-limited resolution, about 180 nm set to conventional microscopies. However, current SR microscopes are largely founded on exogenous fluorescent labelling for biological imaging. Herein, we report label-free stimulated Raman scattering (SRS) microscopy with lateral resolution <110 nm, in which the image contrast originates only from endogenous biomolecules. In the other work, we developed ultrafast 2000 frame/second chemical imaging microscope to visualize the radical polymerization process in action. To achieve multi-colour protein imaging, unnatural amino acid with Raman tags are incorporated into specific protein by genetic code expansion technology. Attributed to the enhancements in spatial resolution, imaging speed, and chemical specificity, we demonstrate broad applications of Raman in biomedical and clinical fields.
