GB 23200.121-2021 食品安全国家标准 《植物源性食品中331种农药及其代谢物残留量的测定 液相色谱—质谱联用法》已正式实施,此标准采用QuEChERS前处理方法、液相色谱-三重四极杆串联质谱一次进样正负源切换同时测定331种农药及其代谢物残留量,解决了现行液质标准适用农产品基质种类少、农药及代谢物品种不全、前处理操作复杂、部分农药方法定量限高于最大残留限量等诸多问题。本文分别以杏鲍菇、上海青、大豆、红茶为植源性样品基质,选取了国标GB23200.121-2021中的164种农药,采用纳谱分析的SelectCore QuEChERS萃取盐包及净化管分别对样品基质进行前处理,使用超高效三重四级杆液相色谱-质谱联用仪进行测定,实验数据表明纳谱分析的SelectCore QuEChERS前处理产品可以有效避免基质杂质干扰,并且实验结果稳定,回收率满足检测要求。

23200.121-2021食品安全国家标准植物源性食品中331种农药及其代谢物残留量的测定液相色谱一质谱联用法,本方法适用于植物源性食品中331种农药及其代谢物残留量的测定。 实验步骤 1、试样制备 样品测定部位按照 GB 2763 附录A 的规定执行。 食用菌、热带和亚热带水果(皮可食)随机取样1 kg, 水生蔬菜、茎菜类蔬菜、豆类蔬菜、核果类水果、热带和亚热带水果(皮不可食)随机取样2 kg,瓜类蔬菜和水果取4~6个个体(取样量不少于1kg),其他蔬菜和水果随机取样3 kg。对于个体较小的样品,取样后全部处理;对于个体较大的基本均匀样品,可在对称轴或对称面上分割或切成小块后处理;对于细长、扁平或组分含量在各部分有差异的样品,可在不同部位切取小片或截成小段后处理;取后的样品将其切碎,充分混匀,用四分法取一部分或全部用组织捣碎机匀浆后,放入聚乙烯瓶中。 干制蔬菜、水果和食用菌随机取样 500 g,粉碎后充分混匀,放入聚乙烯瓶或袋中。 谷类随机取样500 g, 粉碎后使其全部可通过 425 um 的标准网筛,放入聚乙烯瓶或袋中。 油料、茶叶、坚果和香辛料随机取样500 g, 粉碎后充分混匀,放入聚乙烯瓶或袋中。 植物油类搅拌均匀,放入聚乙烯瓶中。 2、试样贮存 将试样按照测试和备用分别存放。于-18℃条件下保存。 3、前处理 3.1蔬菜、水果、食用菌和糖料 称取10g试样(精确至0.01 g) 于50 mL塑料离心管中,加入10 mL乙腈及1颗陶瓷均质子,剧烈震荡1 min, 加入 SelectCore QuEChERS 萃取盐包QS-001(4g无水硫酸镁、1g氯化钠、1g柠檬酸钠二水合物、0.5g柠檬酸二钠盐倍半水合物),剧烈震荡1 min 后4200 r/min 离心5 min。吸取上清液1 mL至 SelectCore QuEChERS 净化管 Q-02P02中 (150 mg无水硫酸镁、25 mg PSA),或吸取上清液6 mL 至 SelectCore QuEChERS 净化管 Q-15P02中(900 mg 无水硫酸镁、150 mg PSA);对于颜色较深的试样,则吸取上清液1 mL 至 SelectCore QuEChERS 净化管 Q-02PG01中(150 mg无水硫酸镁、25 mg PSA、2.5mg GCB), 或吸取上清液6 mL 至含有 GCB 填料的 SelectCore QuEChERS 净化管 Q-15PG01中(885mg无水硫酸镁、150 mg PSA、15 mg GCB),涡旋混匀1 min。 4200 r/min 离心5 min, 吸取上清液过有机系微孔滤膜,待测定。 ( 注:对于干制蔬菜、水果和食用菌,称 取 1g试样(精确至0.01g)于50 mL塑料离心管中,加9 mL水 ) 涡旋混匀,静置30 min 后按上述方式处理。 3.2谷物、油料和坚果 称取5g试样(精确至0.01 g)于50 mL塑料离心管中,加10 mL水涡旋混匀,静置30 min。加入15 mL乙腈-乙酸溶液(量取10 mL 乙酸加入990 mL乙中,混匀)及1颗陶瓷均质子,剧烈震荡1 min, 加入 SelectCore QuEChERS 萃取盐包 QS-002 (6 g无水硫酸镁、1.5 g乙酸钠),剧烈震荡1 min 后4200 r/min 离心5 min。吸取上清液1 mL 至 SelectCore QuEChERS 净化管 Q-02PC02中 (150 mg 无水硫酸镁、50 mg PSA 和 50 mg C18),或吸取上清液 8 mL 至 SelectCore QuEChERS 净化管 Q-15PC01中(1200 mg无水硫酸镁、400 mg PSA 和400 mg C18),涡旋混匀1 min。 4200 r/min 离心5 min, 吸取上清液过有机系微孔滤膜,待测定。 3.3茶叶和香辛料 称取2g试样(精确至0.01 g) 于50 mL塑料离心管中,加10 mL 水涡旋混匀,静置30 min。加入15 mL乙腈-乙酸溶液(量取10 mL 乙酸加入990 mL乙腈中,混匀)及1颗陶瓷均质子,剧烈震荡1 min, 加入 SelectCore QuEChERS 萃取盐包 QS-002(6g无水硫酸镁、1.5g乙酸钠),剧烈震荡1 min 后4200 r/min离心5 min。 吸取上清液1 mL 至 SelectCore QuEChERS 净化管 Q-02PCG03中(150 mg 无水硫酸镁、50 mg PSA、50 mg C18 和 25 mgGCB), 或吸取上清液8 mL 至 SelectCore QuEChERS 净化管 Q-15PCG02中 (1200 mg 无水硫酸镁、400 mg PSA、400 mg C18和200 mg GCB), 涡旋混匀1 min。 4200 r/min 离心5 min, (吸取上清液过有机系微孔滤膜,待测定。 3.4植物油 称取2g试样(精确至0.01g)于50mL塑料离心管中,加入5 mL水。加入10 mL乙腈及1颗陶瓷均质子, 剧烈震荡1 min, SelectCore QuEChERS 萃取盐包 QS-001(4 g无水硫酸镁、1g氯化钠、1g柠檬酸钠二水合物、0.5 g柠檬酸二钠盐倍半水合物),剧烈震荡1 min 后4200 r/min 离心5 min。吸取上清液1mL 至 SelectCore QuEChERS 净化管 Q-02PC02 中(150 mg 无水硫酸镁、50 mg PSA 和50 mg C18), 或吸取上清液8 mL 至 SelectCore QuEChERS 净化管Q-15PC01中(1200 mg无水硫酸镁、400 mg PSA 和 400 mg C18), 涡旋混匀1 min。4200 r/min 离心5 min, 夜取上清液过有机系微孔滤膜,待测定。 注:测定蔬菜、水果、食用菌、糖料、植物油中磺酰脲类除草剂、环己烯烯类除草剂(烯草酮、烯草酮砜、烯草酮亚砜、噻草酮、三甲苯草酮、烯禾啶)、三唑并嘧密磺酰胺类除草剂(双氟磺草胺、唑嘧磺草胺、五氟磺草胺)、氟啶胺、螺虫乙酯及其代谢物、甲磺草胺、苯嘧磺草胺、苯噻隆、氰霜唑代谢物 CCIM 和异噁唑草酮-二酮腈时,PSA 的用量降低至每毫升提取液5 mg, 谷物、油料、坚果降低至每毫升提取液10mg。 1% 50% 3% 实验结论 分别向不同样品空白基质中添加多种农残标准品,杏鲍菇、上海青空白基质中各农药加标量为 0.01 mg/kg,大豆空白基质中各农药的加标量为0.02 mg/kg, 茶叶空白基质中各农药的加标量为 0.05 mg/kg。图2-图5为不同基质的加标总离子流图,图中各平行样品的总离子流图分别用蓝、橙、红、绿四种颜色显示。各各行样本总离子流图轮廓一致,未见偏移,表明不同样品基质样品经过纳谱分析 SelectCore QuEChERS 样品前处理产品处理后农药多残留分析结果稳定。 各农药的加标回收率如上表所示,不同基质样品的加标回收率均能满足检测要求,其中,在杏鲍菇基质中,共137种农药的回收率在 80%-119%之间;在上海青基质中,共128种农药的回收率在 80%-120%之间;在大豆基质中,共136种农药的回收率在 80%-120%之间;在红茶基质中,共149种农药的回收率在80%-120%之间。 本项测定数据由江南大学分析测试中心提供。
