橙汁生产是我国柑橘产业发展的主要动力和方向。大量流行病学和动物试验研究表明, 柑橘类水果具有抗氧化性、抗癌、预防循环系统疾病、抗过敏以及抗菌等作用。这主要是柑橘含有以黄烷酮为主(占80 %) 的类黄酮, 其中橙皮苷(hesperidin) 、柚皮苷(naringin) 是柑橘中最主要的类黄酮。橙皮苷具有抗氧化, 抗炎镇痛、免疫调节、抗肿瘤等作用。柚皮苷在降血脂、镇静、抗氧化、抗肿瘤、抗真菌、抗动脉粥样硬化等方面具有较强的生物活性。随着对橙汁饮料中2 种类黄酮活性成分的检测与含量控制, 对提高橙汁的饮料价值, 引导消费, 促进产业发展具有积极作用。 采用毛细管电泳法可以测定橙皮苷和柚皮苷的含量,是基于饮料的不同成分在毛细管电泳中的分离并测定这些成分在220nm波长下的吸收量来检测这些成分。

HIGH PERFORMANCECAPILLARYELECTROPHORESIS SYSTEM DETERMINATION OF HESPERIDIN AND NARINGIN IN JUICES USINGCAPEL-105M CAPILLARY ELECTROPHORESIS SYSTEM LUMEX Method M 04-67 (2010) INTRODUCTION The method is intended for determination of hesperidin and naringin using capillary electrophoresistechnique in order to reveal adulteration of juices. These components are tested in citrus juices,concentrated juices, nectars, juice-containing beverages, and fruit sauces that are produced primarily fromoranges, grapefruits, tangerines, lemons, and limes. MEASUREMENT METHOD The method is based on dilution of a drink, its subsequent separation, and quantitative determination of thecomponents by capillary electrophoresis. The components are detected by their intrinsic absorption at a wavelength of 220 nm. MEASUREMENT RANGE The measured weight concentrations of the components determined in samples lie in the range20-2000 mg/l. EQUIPMENT AND REAGENTS The following equipment and reagents are used in the measurements: CAPEL-105M capillary electrophoresis system with the high-voltage positive polarity; special cassette with capillary (I.D. 50 um, total length 75 cm); naringin; 0 hesperidin; rectified ethyl alcohol; sodium tetraborate,reagent grade; ● sodium hydroxide, reagent grade; hydrochloric acid, reagent grade. Data acquisition, processing and output areeperformed using a personal computer running underWINDOWS° 2000/XP operating system withinstalled dedicated software for data acquisition andprocessing.
