Many proteins contain, besides prolines in trans configuration, occasionally also proline residues whose conformational equilibrium is shifted to the cis configuration. Studies of these equilibria as well as the influence of cis/trans isomerases are usually performed with peptides as model systems. Using one- and multidimensional proton spectra, or P-NMR, the equilibria ratios can be determined under various experimental conditions. Of special interest is the influence of the isomerases on the protein folding process, which plays an important role in the molecular recognition of biolocially active conformations.
