近期，“天眼问政”栏目对网友关心的“贵阳市面上的桶装饮用水是否符合质检标准”“桶装水饮用是否安全放心”等问题进行调查报道。而后，记者采访了贵州省地质矿产中心实验室/贵州省水质检验检测中心，探访桶装水检测的全过程。pH值、感官指标最先检测记者走进贵州省水质检验检测中心，水质常规仪器室、水质常规分析室、离子色谱室、微生物操作间等各检测功能区合理分布，各实验区域的技术人员忙而有序检测水质。容量法检测水中的酸度及游离二氧化碳据了解，该中心是完全独立于开发、生产、销售方的第三方检测实验室，通过省级实验室资质认定。具备从水源地调查评价、各类水质检验检测到各类水产品的检验检测能力，有一支饮用水检验、水资源评价、咨询的专业队伍作为强有力的技术支撑。根据检测委托需求，贵州省水质检验检测中心实验室通常在一至两周出具检测报告。该中心水质检测技术人员、工程师杜作灵介绍，“收样窗口接受委托，按照产品明示的贮存条件进行妥善保存。样品编码后，根据国家检验检测相关技术要求、检验指标保存的时限要求来安排检测。通常，pH值、感官指标等为急测指标，收样后开瓶最先检测。”记者来到水样库房，做好编码记录等待检验的桶装饮用水避光保存。杜作灵介绍，结合我省优质的天然水源条件，制定了具有贵州特色的《饮用天然泉水》团体标准、地方标准，参考国家矿泉水标准要求，对界限指标未达到国家矿泉水标准的天然优质水源，也相应提出产品特征性界限指标的要求或加严了污染物限量指标的要求等。水样库房，做好编码记录的桶装饮用水避光保存。“天然的水源开采需要经相关管理部门的批准。”杜作灵详解，办理“取水许可证”最严格的要数饮用天然矿泉水，水源需经过严格的水源水质跟踪监测，按采矿的标准办理“采矿许可证”，属于液态的矿产资源。“对水源要求最低的是饮用纯净水和其他饮用水，原料用水可允许使用自来水进行生产。”从水样库房出来，记者参观了水质检测实验室展示的各项检测水质过程。杜作灵说，包装饮用水必要的检测项目主要包含：感官指标、理化指标、污染物限量指标、微生物限量指标。其中，饮用天然矿泉水以及我省特色饮用天然泉水，还需要检测产品的界限指标，如：锶、锌、锂、硒、偏硅酸、溶解性总固体。　桶装水贮存有讲究“通常桶装水没有固定的保质期，桶装水的保质期取决于多种因素，包括贮存条件和水的类型等。一般来说，未开封的桶装水可以保质较长时间，开封后的水应尽快饮用。”该中心科普专职人员、高级工程师贾双琳介绍，未开封的桶装水保质期通常在1个月至3个月不等，瓶装水在1年到2年不等，有些品牌甚至可以更长。水中pH的测定“确切的保质期会因品牌和制造商而异，因此在购买前应先查看标签上的日期，按其标签明示的贮存条件进行妥善保存。”贾双琳建议，开封后的桶装水，水暴露在空气中，可能会受到微生物的污染，桶装水的保质期会缩短。开封后应尽快饮用，以减少细菌滋生的机会。“桶装水的保质期受贮存条件的影响，最好将其存放在干燥、阴凉、避免阳光直射的地方。”贾双琳强调，空气通过饮水机进气孔与水接触，空气中的微生物、藻类孢子等进入水中，特别是营养丰富的矿泉水、天然泉水在开封后，会促使细菌等繁殖生长，缩短保质期。“生活中，我们有时看到桶装水长青苔，这种现象是否说明桶装水变质呢？”记者问。水中挥发性有机物检测的准备工作“有的桶装水会长青苔，因为青苔是藻类的一种，是古老的单细胞低等植物，在日常的办公室和家居环境空气中往往会有微量的藻类孢子存在，特别是在潮湿和靠近绿色植物的地方常有青苔孢子的存在。在光照和适宜的温度条件下，利用水、矿物质、二氧化碳，与其自身的叶绿素进行光合作用，迅速繁殖，从而形成我们肉眼可见的青苔。”贾双琳建议，桶装水要按标签明示的贮存条件进行贮存，保持存放环境的清洁卫生，饮水机要经常清洗。延伸阅读饮用水检测主要指标感官指标主要指人们可以通过观察、闻、品尝等感官方式来判断水质的指标，包括：色度、浊度、状态、滋味气味。　理化指标包括反映水中消毒剂含量的余氯，反映消毒副产物的溴酸盐、三氯甲烷，评估水质安全性的一般化学指标，例如pH、挥发性酚、阴离子合成洗涤剂等，反映放射性污染情况的例如总α放射性、总β放射性指标等。　污染物限量指标反映的是食品从生产过程中产生或由污染环境带入的化学性危害物质，如铅、镉、砷、硝酸盐、亚硝酸盐。微生物限量指标衡量饮用水质量的重要参数之一，包括总大肠菌群、铜绿假单胞菌等。可反映水中是否存在病原微生物的污染，以及水体的清洁程度和污染程度。　界限指标主要反映的是水质健康性指标，是我国评价矿泉水质量最常用、最重要的指标。水中界限指标有锶、锌、锂、硒、偏硅酸、溶解性总固体，如锶能强壮人体骨骼，降低人体对钠的吸收，利于心血管的正常活动；偏硅酸对强壮骨骼、促进生长发育等具有重要意义；硒能提高人体免疫力；溶解性总固体能补充人体对常量组分钾、钠、钙、镁离子的需要，调整人体电解质平衡。

集量5ml,以空腹血为佳，建议使用含有EDTA抗凝剂的真空釆血 管采集血液。 /p p style=" text-indent: 2em " 4.血清标本：尽量釆集急性期、恢复期双份血清。第一份 血清应尽早（最好在发病后7天内）釆集，第二份血清应在发病 后第3～4周釆集。采集量5ml,建议使用无抗凝剂的真空釆血管。血清标本主要用于抗体的测定，从血清抗体水平对病例的感染 状况进行确认。血清标本不进行核酸检测。& nbsp /p p style=" text-indent: 2em " 5．眼结膜标本：出现眼部感染症状的病例需采集眼结膜拭 子标本。 /p p style=" text-indent: 2em " 6．便标本：出现腹泻症状的患者需采集便标本。 /p p style=" text-indent: 2em " strong （四）标本采集方法。 /strong /p p style=" text-indent: 2em " 1．咽拭子：用2根聚丙烯纤维头的塑料杆拭子同时擦拭双 侧咽扁桃体及咽后壁，将拭子头浸入含3ml病毒保存液（也可使 用等渗盐溶液、组织培养液或磷酸盐缓冲液）的管中，尾部弃 去，旋紧管盖。 /p p style=" text-indent: 2em " 2．鼻拭子：将1根聚丙烯纤维头的塑料杆拭子轻轻插入鼻 道内鼻腭处，停留片刻后缓慢转动退出。取另一根聚丙烯纤维 头的塑料杆拭子以同样的方法采集另一侧鼻孔。上述两根拭子 浸入同一含3ml采样液的管中，尾部弃去，旋紧管盖。 /p p style=" text-indent: 2em " 3．鼻咽抽取物或呼吸道抽取物：用与负压泵相连的收集器 从鼻咽部抽取粘液或从气管抽取呼吸道分泌物。将收集器头部 插入鼻腔或气管，接通负压，旋转收集器头部并缓慢退出，收 集抽取的粘液，并用3ml采样液冲洗收集器1次（亦可用小儿导 尿管接在50ml注射器上来替代收集器）。 /p p style=" text-indent: 2em " 4．深咳痰液：要求病人深咳后，将咳出的痰液收集于含3ml 采样液的50ml螺口塑料管中。 /p p style=" text-indent: 2em " 5．支气管灌洗液：将收集器头部从鼻孔或气管插口处插入 气管（约30cm深处），注入5ml生理盐水，接通负压，旋转收集 器头部并缓慢退出。收集抽取的粘液，并用采样液冲洗收集器1次（亦可用小儿导尿管接在50ml注射器上来替代收集）。 /p p style=" text-indent: 2em " 6．肺泡灌洗液：局部麻醉后将纤维支气管镜通过口或鼻经 /p p style=" text-indent: 2em " 过咽部插入右肺中叶或左肺舌段的支管，将其顶端契入支气管 分支开口，经气管活检孔缓缓加入灭菌生理盐水，每次30～50 ml，总量100～250ml，不应超过300ml。 /p p style=" text-indent: 2em " 7．血液标本：建议使用含有EDTA抗凝剂的真空釆血管采集 血液标本5ml，室温静置30分钟，1500～2000rpm离心10分钟， 分别收集血浆和血液中细胞于无菌螺口塑料管中。 /p p style=" text-indent: 2em " 8.血清标本：用真空负压采血管采集血液标本5ml，室温静 置30分钟，1500～2000rpm离心10分钟，收集血清于无菌螺口塑 料管中。 /p p style=" text-indent: 2em " 9．粪便标本：如患者发病早期出现腹泻症状，则留取粪便 标本3-5ml； /p p style=" text-indent: 2em " 10．眼结膜拭子标本：眼结膜表面用拭子轻轻擦拭后，将 拭子头进入采样管中，尾部弃去，悬紧管盖。 /p p style=" text-indent: 2em " 其他材料：依据设计需求规范采集。 /p p style=" text-indent: 2em " strong （五）标本包装。& nbsp /strong /p p style=" text-indent: 2em " 标本采集后在生物安全二级实验室生物安全柜内分装。1．所有标本应放在大小适合的带螺旋盖内有垫圈、耐冷冻的样本采集管里，拧紧。容器外注明样本编号、种类、姓名及 采样日期。 /p p style=" text-indent: 2em " 2．将密闭后的标本放入大小合适的塑料袋内密封，每袋装一份标本。样本包装要求要符合《危险品航空安全运输技术细 则》相应的标准。 /p p style=" text-indent: 2em " 3. 涉及外部标本运输的，应根据标本类型，按照A类或B类 感染性物质进行三层包装。 /p p style=" text-indent: 2em " strong （六）标本保存。 /strong /p p style=" text-indent: 2em " 用于病毒分离和核酸检测的标本应尽快进行检测，能在24 小时内检测的标本可置于4℃保存；24小时内无法检测的标本则 应置于-70℃或以下保存（如无-70℃保存条件，则于-20℃冰箱 暂存）。血清可在4℃存放3天，-20℃以下可长期保存。应设立 专库或专柜单独保存标本。标本运送期间应避免反复冻融。 /p p style=" text-indent: 2em " strong （七）标本送检。 /strong /p p style=" text-indent: 2em " 标本采集后应尽快送往实验室，如果需要长途运输标本， 建议采用干冰等制冷方式进行保藏。 /p p style=" text-indent: 2em " 1．上送标本。各省（自治区、直辖市）聚集性病例的标本，上送中国疾病预防控制中心病毒病预防控制所进行检测复核，并附样本送检单（见附件）。 /p p style=" text-indent: 2em " 2．病原体及标本运输。 /p p style=" text-indent: 2em " 2.1国内运输。新型冠状病毒毒株或其他潜在感染性生物材料的运输包装分类属于A类，对应的联合国编号为UN2814，包装符合国际民航 组织文件Doc9284《危险品航空安全运输技术细则》的PI602分类包装要求；环境样本属于B类，对应的联合国编号为UN3373， 包装符合国际民航组织文件Doc9284《危险品航空安全运输技术 细则》的PI650分类包装要求；通过其他交通工具运输的可参照 以上标准包装。 /p p style=" text-indent: 2em " 新型冠状病毒毒株或其他潜在感染性材料运输应按照《可 感染人类的高致病性病原微生物菌（毒）种或样本运输管理规 定》（原卫生部令第45号）办理《准运证书》。 /p p style=" text-indent: 2em " 2.2国际运输。新型冠状病毒毒株或样本在国际间运输的，应规范包装，按照《出入境特殊物品卫生检疫管理规定》办理相关手续，并 满足相关国家和国际相关要求。 /p p style=" text-indent: 2em " 2.3毒株和样本管理。新型冠状病毒毒株及其样本应由专人管理，准确记录毒株 /p p style=" text-indent: 2em " 和样本的来源、种类、数量，编号登记，采取有效措施确保毒 株和样本的安全，严防发生无用、恶意使用、被盗、被抢、丢 失、泄露等事件。 /p p style=" text-indent: 2em " strong 二、新型冠状病毒的实验室检测 /strong /p p style=" text-indent: 2em " 新型冠状病毒感染的常规检测方法是通过实时荧光RT-PCR 鉴定。任何新型冠状病毒的检测都必须在具备适当条件的实验 室由经过相关技术安全培训的人员进行操作。本指南中的核酸 检测方法主要针对新型冠状病毒基因组中开放读码框1ab（open reading frame 1ab ， ORF1ab ） 和 核 壳 蛋 白 （ nucleocapsid /p p style=" text-indent: 2em " & nbsp protein，N）。 /p p style=" text-indent: 2em " 在实验室要确认一个病例为阳性，满足以下条件：同一份标本中新型冠状病毒2个靶标（ORF1ab、N）特异性实时荧光RT-PCR检测结果均为阳性。如果出现单个靶标阳性的 检测结果，则需要重新采样，重新检测。 /p p style=" text-indent: 2em " & nbsp /p p style=" text-indent: 2em " 阴性结果也不能排除新型冠状病毒感染，需要排除可能产生假阴性的因素，包括：样本质量差，比如口咽等部位的呼吸 道样本；样本收集的过早或过晚；没有正确的保存、运输和处 理样本；技术本身存在的原因，如病毒变异、PCR抑制等。 /p p style=" text-indent: 2em " strong 三、实时荧光 RT-PCR 方法检测新型冠状病毒核酸 /strong /p p style=" text-indent: 2em " strong （一）目的。 /strong /p p style=" text-indent: 2em " 规范实时荧光RT-PCR方法检测新型冠状病毒核酸的工作程 序，保证实验结果的正确可靠。 /p p style=" text-indent: 2em " strong （二）范围。 /strong /p p style=" text-indent: 2em " 适用于实时荧光RT-PCR方法检测新型冠状病毒核酸。 /p p style=" text-indent: 2em " strong （三）职责。 /strong 检测人员：负责按照本检测细则对被检样本进行检测。复核人员：负责对检测操作是否规范以及检测结果是否准确进行复核。 /p p style=" text-indent: 2em " 部门负责人：负责对科室综合管理和检测报告的审核。 /p p 注：如果用的是商品化试剂盒，则以厂家提供的说明书为准。 /p p style=" text-indent: 2em " strong 四、病原生物安全实验活动要求 /strong /p p style=" text-indent: 2em " 根据目前掌握的新型冠状病毒的生物学特点、流行病学特征、临床资料等信息，该病原体暂按照病原微生物危害程度分 类中第二类病原微生物进行管理，具体要求如下： /p p style=" text-indent: 2em " strong （一）病毒培养。 /strong /p p style=" text-indent: 2em " 病毒培养是指病毒的分离、培养、滴定、中和试验、活病 毒及其蛋白纯化、病毒冻干以及产生活病毒的重组实验等操作。上述操作应当在生物安全三级实验室的生物安全柜内进行。使 用病毒培养物提取核酸，裂解剂或灭活剂的加入必须在与病毒 培养等同级别的实验室和防护条件下进行，裂解剂或灭活剂加 入后可比照未经培养的感染性材料的防护等级进行操作。实验 室开展相关活动前，应当报经国家卫生健康委批准，取得开展 相应活动的资质。 /p p style=" text-indent: 2em " strong （二）动物感染实验。 /strong /p p style=" text-indent: 2em " 动物感染实验是指以活病毒感染动物、感染动物取样、感 染性样本处理和检测、感染动物特殊检查、感染动物排泄物处 理等实验操作，应当在生物安全三级实验室的生物安全柜内操 作。实验室开展相关活动前，应当报经国家卫生健康委批准， 取得开展相应活动的资质。 /p p style=" text-indent: 2em " strong （三）未经培养的感染性材料的操作。 /strong & nbsp /p p style=" text-indent: 2em " 未经培养的感染性材料的操作是指未经培养的感染性材料 在采用可靠的方法灭活前进行的病毒抗原检测、血清学检测、 核酸提取、生化分析，以及临床样本的灭活等操作，应当在生 物安全二级实验室进行，同时采用生物安全三级实验室的个人 防护。 /p p style=" text-indent: 2em " strong （四）灭活材料的操作。 /strong /p p style=" text-indent: 2em text-align: left " 感染性材料或活病毒在采用可靠的方法灭活后进行的核酸 检测、抗原检测、血清学检测、生化分析等操作应当在生物安 全二级实验室进行。分子克隆等不含致病性活病毒的其他操作， 可以在生物安全一级实验室进行。 /p p style=" text-indent: 2em text-align: center "
