    Counting, Computing and Reporting Results
    Optimal colony density per filter is 20 to 200. Report all colonies counted as colony forming-units
    (CFU)/mL. Include in the report the method used, the incubation temperature and time, and
    the medium.
    For example: 98 CFU/L, mL, 35 °C, 24 hours, m-HPC broth.
    1 to 2, or fewer colonies per square — Count all of the colonies on the filter, and divide the
    results by the volume of original sample used.
    For example, if there are 122 colonies on the filter, and the volume of original sample used was 10
    mL, compute results as follows:
    122 colonies
    ---------------------- = 12.2 CFU/mL
    10 mL sample

    3 to 10 colonies per square — Count all colonies in 10 representative squares and divide by 10
    to obtain an average number of colonies per square. Multiply this number by 100 and divide by the
    volume of original sample used.
    For example, if you calculated an average of 8 colonies per square, and the volume of original
    sample used was 0.1 mL, compute results as follows:
    8 colonies/square x 100
    ------------------------------------ = 8000 CFU/mL
    0.1 mL sample

    10 to 20 colonies per square — Count all colonies in 5 representative squares and divide by 5 to
    obtain an average number of colonies per square. Multiply this number by 100 and divide by the
    volume of original sample used.
    For example: if there are an average of 17 colonies per square, and the volume of original sample
    used was 0.1 mL, compute results as follows:
    17 colonies/square x 100
    ------------------------------------- = 17, 000 CFU/mL
    0.1 mL sample

    g) Count colonies on membrane filters at 10 to 15 x magnification.

    Preferably place petri dish on microscope stage slanted at a 45° angle

    and adjust the light source vertical to the colonies. Optimal colony

    density per filter is 20 to 200. If colonies are small and there is no

    crowding, a higher limit is acceptable. Count all colonies on the

    membrane when there are 1 to 2, or fewer, colonies per square. For 3

    to 10 colonies per square count 5 squares and obtain average count

    per square. Multiply average count per square by 100 times the

    reciprocal of the dilution to give colonies per millilitre. If there are more

    than 20 colonies per square, record count as >2000 times the

    reciprocal of the dilution. Record counts only when there are discrete

    separate colonies without spreaders.

    在这里square应该就是指“方格”,看下面的图示:



    从上面我附的这篇文章来看,菌落数的计算方式有以下三种:

    1 to 2, or fewer colonies per square —Count all of the colonies on the filter, and divide the

    results by the volume of original sample used.

    如果一个方格上的菌落数为1-2个或者更低,则直接将滤膜上所有菌落数的总和除以取样体积即得测试结果(单位是CFU/ml)。

    3 to 10 colonies per square —Count all colonies in 10 representative squares and divide by 10 to obtain an average number of colonies per square. Multiply this number by 100 and divide by the volume of original sample used.

    如果一个方格上的菌落数为3-10个,则取10个有代表性的方格进行计算。

    10 to 20 colonies per square —Count all colonies in 5 representative squares and divide by 5 to obtain an average number of colonies per square. Multiply this number by 100 and divide by the volume of original sample used.

    如果一个方格上的菌落数为10-20个,则取5个有代表性的方格进行计算。

    当方格上的菌落数为1-2个或者更低时,将滤膜上的所有菌落计数

    这种方式通常应该是不断地稀释直至一个方格上的菌落数为1-2或更低

    我也没有用这种方法计数过,对这方面不太了解,这也是我感到比较纳闷的地方

    首先非常感谢几位老师的热心帮助!其实昨天看到poorlittle老师说square是可能指“方格”时,自己仔细琢磨后感觉对那句话有些小明白,只是不能完全确定,因为之前译成“平方”,但一直感觉那样译不通顺,现在影子老师讲的如此详细,终于明白了。
    在这里很想说这个论坛真是个好论坛,既能得到大家的帮助,又让人学到好多专业的英语知识,要多多支持!!
