3.1.4 巯基和二硫键定位 二硫键在维持蛋白和多肽三级结构和正确折叠中具有重要
作用,同时也是研究翻译后修饰所经常面临的问题,自由巯基在研究亚基之间及蛋白与
其他物质相互作用中具有重要意义。利用碘乙酰胺、4-乙烯吡啶、2-巯基苏糖醇等试剂
对蛋白质进行烷基化和还原烷基化,结合蛋白酶切、肽谱技术,利用生物质谱的准确分
子量测定,可实现对二硫键和自由巯基的快速定位与确定[11]。这在含有多二硫键结构
的活性多肽与蛋白质研究中有重要用途。
3.1.5 蛋白质翻译后修饰 蛋白质在翻译中或翻译后由于不同功能所需会发生二十多
类修饰,其中最常见的是磷酸化和糖基化,这些修饰将影响蛋白质的电荷数、疏水性、
构象和稳定性,进而影响其生物活性,因此在基因工程重组蛋白评估时,翻译后修饰也
被认为是影响活性的一个重要因素。传统的Edman降解技术会破坏蛋白质修饰,肼解方法
虽能得到糖链并鉴定之,但却不能进行糖链的准确定位。结合肽谱和脱磷酸酶作用,目
前已可以用MALDI-TOF-MS 对双向电泳分离蛋白质磷酸化位点进行定位[12]。凝集素可对
糖链进行特异性吸附,利用这一性质,可用凝集素对糖蛋白酶解后的肽混合物进行选择
性吸附,MALDI-TOF-MS测定质量数,再用糖苷酶降解含糖肽,质谱测定后即可确定糖链
的大小,结合不同的酶解方式可确定糖基化位点[13],选择不同的凝集素,还可确定糖链
的连接方式。串联质谱技术中子离子扫描模式可以快速选择被修饰片段,然后可根据特
征丢失确定修饰类型[14],是目前最有效的对蛋白质翻译后修饰进行识别与鉴定的分析
手段。这方面的研究已有大量文献报道。鉴于修饰的多样性,目前已有人结合生物质谱
技术,通过分析数千例蛋白质翻译后修饰,建立蛋白质修饰数据库FindMod[15],其完善
和发展必将推动蛋白质大规模鉴定的进程。
3.1.6 生物分子相互作用及非共价复合物 蛋白质与其他生物分子相互作用在信号传
导、免疫反应等生命过程中起重要作用。软电离技术的发展,促进了生物质谱在蛋白质
复合物研究中的应用,目前已涉及了分子伴侣对蛋白折叠作用、蛋白/DNA复合物、RNA/
多肽复合物、蛋白质-过渡金属复合物及蛋白-SDS加合物等多种类型的复合物的结构及结
合位点的研究[16~20]。
3.2 多糖结构测定
多糖的免疫功能是近年来研究的热点领域,其结构的测定是功能研究的基础。多糖
不像蛋白质和核酸,其少数的分子即可由于连接位点的不同,而形成复杂多变的结构,
因而难以用传统的化学方法研究。生物质谱具备了测定多糖结构的功能,配以适当的化
学标记或酶降解,可对多糖结构进行研究[21]。采用MALDI-TOF-MS已对糖蛋白中的寡糖
侧链进行了分析,包括糖基化位点、糖苷键类型、糖基连接方式以及寡糖序列测定[22]
等。与传统的化学方法相比,质谱技术具有操作简便、省时、结果直观等特点。
3.3 寡核苷酸和核酸的分析
目前,生物质谱已经实现对数十个碱基寡核苷酸的分子量和序列测定[23],此技术
可用于天然或人工合成寡核苷酸的质量控制。人类基因组有30亿个碱基,但真正与疾病
有关的只是少数可变的基因。基因库中有一个很丰富的资源即300万个单核苷酸多态性片
段(SNPs),它是一类基于单碱基变异引起的DNA多态性,由于其位点丰富(约一个多态
性每一千个碱基),使得在鉴定和表征与生物学功能和人类疾病相关的基因时,它可作
为关联分析的基因标志。SNPs不一定要准确定位,关键是测定其在种群中出现的频率及
其遗传和表型的关系,这便需要高通量的测定技术。生物质谱可以通过准确的分子量测
定,确定SNP与突变前多态性片段分子量差异,如碱基由A突变为C后,SNP分子量增加24
.025,突变为G时,增加15.999,由分子量的变化可推定突变方式。一种快速而经济的方
法是利用目前不断成熟的DNA芯片技术和质谱检测相结合,将杂交至固定化DNA阵列上的
引物进行PCR扩增后,直接用质谱对芯片上SNP进行检测,该法将所需样品的体积由微升
减至纳升,且有利于自动化和高通量的测定[24]。Griffin等用浸入剪切分析(一种不经
PCR而可以直接进行SNPs分析的信号放大方法)结合MALDI-TOF-MS分析人基因组SNPs,该
法既节省时间,又适于高通量分析,有利于特异性基因的定位、鉴定和功能表征[25]。
DNA在内环境中的温度、pH、机体代谢过程产生的超氧化物自由基、外环境中的各种
化学物质(如烷化剂)、物理因素(紫外线等)各种条件作用下,都可能发生损伤,若
损伤不能及时修复,就会产生严重的生物学后果。DNA损伤不仅包括DNA分子中的碱基,
而且还包括脱氧核糖和磷酸,其中碱基损伤最厉害,造成的后果也最严重。碱基错配、
电离辐射导致串联损伤、活性氧损伤、过渡金属Cr(Ⅲ)复合物与DNA作用引发的损伤以及
致癌剂对DNA损伤等的生物质谱研究多有报道[26~30]。
3.4 药物代谢
近年来质谱在药物代谢方面的研究进展迅速。其主要研究药物在体内过程中发生的
变化,阐明药物作用的部位、强弱、时效及毒副作用,从而为药物设计、合理用药提供
实验和理论基础。特别是采用生物技术获得的大分子药物的体内代谢研究,更是传统的
研究手段难以解决的难题。体内药物或代谢物浓度一般很低,而且很多情况下需要实时
检测,而质谱的高灵敏度和高分辨率以及快速检测则为代谢物鉴定提供了保证。LC-ESI
-MS-MS在这方面有独特的优势,由于对液态样品和混合样品的分离能力高,可通过二级
离子碎片寻找原型药物并推导其结构,LC-ESI-MS-MS已广泛地应用于药物代谢研究中一
期生物转化反应和二期结合反应产物的鉴定、复杂生物样品的自动化分析以及代谢物结
构阐述等[31,32]。
3.5 微生物鉴定
微生物的检验在环境监测、农产品分析、食品加工、工业应用、卫生机构维护及军
事医学中都很重要,其重点主要在于微生物的分类鉴定上。由于微生物成分一般不是特
别复杂,目前的ESI和MALDI技术已可以在其全细胞水平展开。
对微生物全细胞蛋白成份的鉴定,可用MALDI-MS或ESI-MS对裂解细胞直接检测,测
定其全细胞指纹谱,找出种间和株间特异保守峰作为生物标记(biomarker)[33],以此
来进行识别。美国军方已经开展这方面的研究,其目的是通过大量指纹谱的测定,构建
数据库,以实现对细菌等微生物的快速鉴定。细菌鉴定的另一个主要方法是细菌的脂肪
酸图,即细菌脂类水解后释放出脂肪酸,将这些脂肪酸甲基化后分离检测所得到的图谱
。传统上用气相色谱(GC)分离,火焰离子检测器检测,速度慢且麻烦,质谱软电离技
术的出现大大提高了其鉴定速度。有报道用热裂解甲基化法结合离子肼质谱来区分20多
种细菌[34]。除了生物标记和脂肪酸作图外,也有人用细菌糖类化合物作图,来作为细
菌鉴定的依据,同时也用它来描述细胞所处的生理状况[35]。
生物除污(bioremediation)是利用微生物把污染物转换为低害或无害物。特异降解
微生物的选择及其代谢性能的鉴定是该技术的关键。MALDI-TOF-MS多次被用于监测细菌
的降解能力以及在外界刺激条件下细菌蛋白质组的变化[36,37]。
3.6 其他应用
质谱在医用材料如人造血、新型生物假肢、人造皮肤等研究中也有较大的潜在用途
。最主要的是分析检测这些材料中活性物质的纯度、浓度及结构等,这是生物材料质量
控制的重要依据之一。
